Typical mixed cultures of T. aceti contain few large organisms, only about 15% being above the screen cutoff size. There is a 2- to 3-day safety factor after screening before reproduction begins. Therefore, the screened cultures are allowed to incubate for at least 24 hr before FUdR is added. A suitable concentration, using the defined medium, is 100 μg/ml of FUdR plus 150 ^g/ml of uridine. In Roux bottles, each containing 50 ml of medium at 30°C, the organisms grow to an average size of 1400 μπ\ in 13 days, the majority being between 1200 and 1600 μ,ιη.

If a greater concentration is used, the organisms simply die back to this level. On the assumption that the difficulty was related to lack of oxygen (although this is not a problem in non-aging cultures), the cultures were shaken during aging. The results were unsatisfactory since the young organisms, although they survive well, do not grow in size. Turbatrix aceti, under normal conditions, does not reproduce well when shaken (Hieb and Rothstein, 1975). Hieb and Rothstein (1975) found that the half-life of cultures grown in Roux bottles is 26 days, in good agreement with the 25 days reported by Gershon (1970).
